Chasing a target of 155 runs, Peshawar Zalmi comfortably reached the total in the 19th over with only two wickets down.
Captain Babar Azam played a match-winning unbeaten innings of 71 runs off 51 balls, while Mohammad Haris contributed 35 runs.
Earlier, Quetta Gladiators were bowled out for 155 runs, with Hasan Nawaz scoring 37 and Rilee Rossouw adding 26.
For Peshawar, Sufiyan Muqeem and Basit Ali took three wickets each.
